Emily Rose universitetdə oxumaq üçün kənddəki təhlükəsiz evini tərk edəndə onu nələrin gözlədiyindən xəbərsiz idi. Bir gecə yataqxanada tək olarkən dəhşətli bir 'hallüsinasiya' görür və huşunu itirir. Bu tutmalar daha tez-tez və ciddi hal almağa başlayanda, dindar bir katolik olan Emily kilsəsinin keşişi Ata Richard Moore-dan ona şeytan çıxarma ayini etməsini istəyir. Gənc qız bu dəhşətli ayin zamanı dünyasını dəyişdikdə, keşiş səhlənkarlıq nəticəsində ölümə səbəb olmaqda ittiham olunaraq məhkəməyə verilir.
