Tehdit filmi günümüzdən 30 il əvvəl, Afrikada başlayır. Amerikalı həkimlər ölümcül bir virusun mühasirəyə aldığı qəsəbə sakinlərini xilas etməyə çalışarkən, yüksək rütbəli məmurlardan birinin verdiyi əmrlə orada olan hər kəs öldürülür. Müdafiə nazirliyinin kökündən həll etdiyini düşündüyü problem illər sonra yenidən baş qaldırır və dolayısı yollarla Yeni Dünyaya daxil olan virus bir anda amerikalıları əsir alır. Hadisəni həll etmək isə yeni boşanmış cütlük olan Sem Deniels (Dastin Hoffman) və həkim Robbi Kio (Rene Russo)-nun öhdəsinə düşür.
