Tarixinin ən məşhur macəraçı arxeologu Indiana Jones'a atası Dr. Henry Jones tərəfindən bir gün kitab göndərilir. Bu kitabın içərisində sirli Müqəddəs Kasa ilə bağlı bəzi ipuçları və bir xərita vardır. Xüsusi bir kolleksionerdən atasının itkin düşdüyünü öyrənən Indiana Jones, muzey kuratorı Marcus Brody'ni də özü ilə götürərək İtaliaya atasını axtarmağa gedir. Burada özlərini tapdıqları yer isə bir Nazi ərazidir və Nazilərin də Müqəddəs Kasanın üzərində göz dikilmişdir. Atasını qurtarmağa gedən Indiana'nın birinci məqsədi Müqəddəs Kasanı Nazilərdən əvvəl əldə etmək olacaqdır.
